Immunohistochemistry (Formalin/PFA-fixed paraffin-embedded sections) - Anti-GFAP antibody (ab4674) IHC image of GFAP staining in a formalin-fixed, paraffin-embedded mouse normal brain tissue section. GFAP (Glial Fibrillary Acidic Protein) is a member of the class III intermediate filament protein family. It is heavily and specifically expressed in astrocytes and certain other astroglia in the central nervous system, in satellite cells, in peripheral ganglia and in non-myelinating Schwann cells in peripheral nerves. Monoclonal Anti-Glial Fibrillary Acidic Protein (GFAP) antibody produced in mouse is suitable for immunohistochemistry at a working dilution of 1:400 using rat brain sections (alcohol-fixed) and microarray analysis. It may be used for immunocytochemical localization of GFAP in human, pig, and rat tissues. Monoclonal antibody is produced by immunizing animals with native GFAP purified from pig spinal cord.

Background The cytoskeleton consists of three types of cytosolic fibers: microfilaments (actin filaments), intermediate filaments, and microtubules. The cells were fixed with 4% paraformaldehyde for 15 min at room temperature, and then permeabilized and blocked with 0.25% Triton X-100 and 10% donkey serum in PBS for 20 min. Samples were then incubated with a Mouse GFAP monoclonal antibody (green; Product # MA5-12023) at a dilution of 1:500 in PBS containing 0.25% Triton X-100 and 10% donkey Western blot analysis of GFAP in various whole cell extracts using GFAP polyclonal antibody (Product # PA5-85261) using 30 µg of sample at a dilution of 1:20000. Sample was then incubated with HRP-conjugated anti-rabbit IgG secondary antibody. Prior to incubation with primary antibody, the sample was separated on 10% SDS-PAGE. Cells were stained with a GFAP mouse monoclonal antibody (Product # MA5-12023) at a dilution of 1:200 in 3% BSA in PBS for 1 hr at RT, and then incubated with Invitrogen™ AlexaFluor™ 488 Plus goat anti-mouse IgG secondary antibody (Product # A32723) at a dilution of 1:1000 for 1 hr at RT. Immunofluorescent analysis of 4% Paraformaldehyde-fixed, 0.1% TritonX-100 permeabilized mouse primary neural mix culture cells labelling GFAP with ab68428 at 1/250 dilution, followed by ab150077 AlexaFluor ® 488 Goat anti-Rabbit secondary antibody at 1/1000 dilution (Green).
