Frustrated with agarose gel electrophoresis? SHIMADZU

The fluorescence of EtBr increases 21-fold upon binding to double-stranded RNA and 25-fold on binding double-stranded DNA so that destaining the background is not necessary with a low stain concentration (10 μg/ml). A trypanocidal agent and possible antiviral agent that is widely used in experimental cell biology and biochemistry. Ethidium has several experimentally useful properties including binding to nucleic acids, noncompetitive inhibition of nicotinic acetylcholine receptors, and fluorescence among others. It is most commonly used as the bromide. Ethidium bromide is a sensitive fluorescent stain for visualizing DNA in agarose and polyacrylamide gels.

Ethidium bromide is not a regulated hazardous material, but due to its potential health hazards, precautions should be taken during handling and disposal. Ethidium bromide gels, stock solutions, buffers and contaminated debris should be referred to EHS for collection and disposal. The mechanism of the enhancement of the fluorescence of ethidium bromide on binding to double helical RNA and DNA has been investigated.

Description Ethidium bromide is a sensitive fluorescent stain for visualizing DNA in agarose and polyacrylamide gels. It is excited with a standard 302 nm UV transilluminator and emits a red-orange signal that can be photographed with Polaroid film or with a CCD-based gel documentation system.

Ethidium bromide

Ethidium bromide has been used in a number of fluorimetric assays for nucleic acids.15,16,17 It has been shown to bind to singlestranded DNA (although not as strongly)2 and triplestranded DNA.18 Because of the binding to DNA, EtBr is a powerful inhibitor of DNA polymerase.4 E7637, Molecular Biology grade powder, is suitable for use in gel electrophoresis and DNA isolation procedures. Ethidium bromide possesses UV absorbance maxima at 300 and 360 nm.
